The disk could be new, as yours is, or it could be just a problem with the download of those large files, which are first always stored once in a Temp directory, then when saved are moved to a permanent directory, then when installed are decompressed to an even larger filesize first and then placed in yet another directory. Have used that machine both for testing and getting work done for quite a few years and at times have gotten the very same problem - skipping or even missing audio due to bad loading of Realtracks. The song will now take a long time to generate before play is pressed (10-30 seconds), but playback will likely be glitch free. If you have a song with lots of RealTracks that is causing problems, press the Song Settings button, and select "This song has playback problems, disable fast generation". If its an old, slow PC, there are things you can do. My PC is 3,500 and runs real well with BiaB. Really fast new machines are around 8,000 speed. Minimum would be geekbench= 1,100, although in the 1,100 to 1,500 range things will work slower, and you could get hiccups with certain songs, other programs running etc. You should run Geekbench to get an overall score of your system speed. Note that I have changed or set many menu short cuts (see BioEdit stuff to change after installation below) to make things quicker, thus these instructions are based on these changes. Once I have edited all of my chromatograms I copy the. The reason why I paste them to a new file first is that importing from the clipboard (File, Import from Clipboard) will place them at the bottom of your file, which is usually not where I want them be. I then select those sequences (control-shift-a), cut (control-shift-c) or copy them (control-a) and paste them (control-s) to the desired BioEdit file. In the other copy of BioEdit I usually go File, New from Clipboard. If you need to copy and paste between copies of the program select the sequences, go Edit, Copy Sequences to clipboard (FASTA Format). The regular copy and paste features work between copies of the program, but copying and pasting sequences does not. One quirk of BioEdit is that if you double click a data file it will open in a new copy of BioEdit, not in an existing one. I would recommend saving everything in fasta format since that is the format I use in order to convert the data to another format or to another person (who probably doesn’t have a copy of BioEdit). As far as I can tell there is no difference between saving your file as a BioEdit formatted file versus as a fasta file. I use this feature on nearly every dataset I create. It is the only program I know of that allows you to edit, search and replace, and paste over the sequence title names independent of your sequences. Aside from that limitation (which isn’t as bad as it might sound once you learn a few tricks), I really like its features. The most annoying aspect is that you have to manually align up each sequence and manually create a consensus sequence (which commercial programs like Sequencher and Geneious are very good at). I use BioEdit to align sequences as it is free and has some handy features. Each of the commercial versions have a free 60 day trial should you wish to try them. I’ve always used the free Chromas version, Chromas Lite, but there are two other versions with more features that are fairly cheap. One trick I find useful later is to always edit your sequences from the same starting base (unless the starts are all messy), as it makes sequence alignment much easier later. However, all things being equal, we do have a sense of how long certain crops should last in cold storage. But how long can seeds really stay viable up there? Should anything disastrous occur to an institution itself, the seeds will still be protected in Svalbard and can be resupplied to the seed bank that experienced the loss. The new sound was steeped in hipster jazz, so in Europe, the 1975 album was given a lively, pop-art cover by artist Jacques Moitoret that harked back to the hippest corners of the early 60s. Gazeuse! was an important transitional album for Gong, who’d lost visionary leader Daevid Allen an album earlier and were now dispensing with vocals altogether. Gong – Gazeuse! Having already powered through a run of Queensland shows, Rudd is currently in the midst of his trek through New South Wales, with this week seeing him visit the likes of Sydney and Newcastle before dipping down to the nation’s capital for a show at the Canberra Theatre. Joining him on his tour is First Nations duo Marlon x Rulla, whose acclaimed performance at the 2021 edition of WOMADelaide saw Rolling Stonenote that, “the group not only cemented themselves as one of the most monumental names on the bill for the evening, but set themselves aside as a pair of artists that you not only should, but undoubtedly need, to pay attention to as their career continues to rise.” After wrapping up locally, Rudd will head overseas for his international tour taking in 18 countries across 48 dates with shows already sold out in the likes of Antwerp, Amsterdam and Belgium. Kicking off his run of dates back in late May, the extensive national trek sees Xavier Rudd making an absolutely mammoth 86 stops, visiting every state and territory in Australia across 38 dates as he traverses the expansive natural landscape that inspires his work. But, it’s not enough for Rudd to simply take his new work on the road with a run of headline dates in major cities (including Perth, Adelaide, Darwin, and Hobart, of course), rather, he’s an artist who doesn’t do things by half measures, having announced a truly immense national tour. With the record out in the world now, the next logical step for Xavier Rudd was to hit the road to share with his adoring fans the breathtaking songs that make up his latest album. “Weaving together organic sounds of nature and wildlife, themes of environmentalism and Indigenous history-and powerful compositions that sees him working with the likes of J-MILLA or the Slabb Brothers- Jan Juc Moon is one of Xavier Rudd ‘s most resonant albums to date,” wrote Rolling Stone of the record back upon its release. His seventh consecutive album to place in the ARIA top ten, the record showcases the arresting quality of Rudd’s passion and musicianship, intertwining in a way that can only be described as magical.
